vista not booting up

Some 12 months ago my Medion MD8833 desktop machine would not start up in the morning, I had to switch off at the back with the switch on the power supply, and this was time sensitive. It would not enter the BIOS until it had been off for 10 or more hours, I left it switched on for about 10 months but eventually I re-installed Vista and it was able to restart in the mornings O/K. Until then it had been off only during restarts for updates. Initially I approached a computer repair shop and after some discussion it was suggested that the power supply appeared to be the key component to check, however I was reluctant to give it to someone I did not know, competence wise, I carried on with machine being left switched on all the time. Since reloading Vista and only 'essential programmes' it has been fine and is switched off overnight and in the morning switched on O/K. Until this morning, it would not switch on as normal, I switched off at the power supply and left it for a couple of hours, when I switched on it reached the password screen and it seemed alright, during this time I used my netbook. I went back to the desktop and it had crashed and now it will not enter the BIOS. I again feel the power supply could be the key component to check - how do I start? I have basic tools and a multimeter, I have considered removing all the usb items and extra memory initially in case there is a problem there. Is the a basic check list for voltage checks?
